Particle size and flow
Lactohale® and Respitose® sieved lactose grades are pharmaceutical alpha-lactose monohydrate with a controlled particle size. The particle size distribution is typically relatively narrow and the median particle size is typically larger than for milled lactose. The Carr's Index indicates fair to good flow. Sieved lactose is therefore beneficial for formulations where flow is of key importance. This lactose type is for example typically used in reservoir devices. Good flow and consistent density of the powder bed ensures the dosing unit is always filled with a constant amount of powder. Sieved lactose grades are also recommended in other platforms when powder flow is required, for example in dosing disk capsule filling technology. More cohesiveness can be obtained by using milled lactose powders.
Besides required flow in the device and filling platform, the particle size of inhalation grade lactose is typically chosen to optimize the drug deposition. By using a combination of different lactose grades, the flow and fines content of the formulation can be steered towards optimal product performance.
